RNA Interference and siRNA

RNA Interference is a mechanism for RNA-guided regulation of gene expression in which double-stranded ribonucleic acid inhibits the expression of genes with complementary nucleotide sequences. It is a naturally occurring cellular mechanism and can be used by researchers to specifically target and silence any gene of interest.

After Tuschl et al's keynote paper in Cell, 2000 on how 21-23mer double-stranded RNA (dsRNA) directs the sequence-specific degradation of mRNA leading to specific gene knockdown, a wave of research emerged using short interfering RNA (siRNA) and RNAi as a tool to study Functional Genomics, Developmental Biology, Gene Expression and in vivo and in vitro Transfection Efficiency studies.

There are various approaches to study RNAi, synthetic siRNAs and shRNA constructs are 2 of the numerous options available. 1st BASE provides the custom synthesis of sequence-specific siRNA for direct transfection, as well as the routine synthesis of shRNA oligos, ~60 bases long purified DNA oligos to be cloned to form shRNA constructs. Our siRNA duplexes will be HPLC purified, and annealed before delivery. A standard siRNA duplex order (5 OD yield) will be delivered 7 working days upon order confirmation. Chemically modified siRNA duplexes, such as fluorescent 6-FAM labels are also available. Prices will be valid for duplexes 19-23-mers in length.

In order to conduct a quantifiable knockdown experiment, it is crucial to set up control reactions (positive, negative and transfection controls). 1st BASE has a comprehensive range of ready-made validated siRNA controls to help researchers set up their controls with ease!
