Sample Preparation Requirements for International Customers

DNA Sequencing Sample Preparation - PCR Clean-up (SS1012, SS1013)
We accept PCR samples straight off the thermal cycler for template DNA purification. Samples will undergo a purification step using a silica PCR clean-up column kit. We accept either individual PCR reactions in single tubes or 96-well plate format.
DNA Sequencing Sample Preparation - Gel Extraction by General Agarose (SS1014, SS1015)
This is similar to the PCR Clean-up described above. Instead of purifying the PCR mix straight off the PCR reaction, they are first run on a gel. Then the target band is excised and DNA extracted using a silica gel extraction column kit.
DNA Sequencing Sample Preparation - Gel Extraction by PAGE (SS1016)
Similar to DNA Sequencing Sample Preparation – Gel Extraction (SS1014, SS1015) except that the medium for analysis is with polyacrylamide gels instead of agarose gels.
DNA Sequencing Sample Preparation - Plasmid Extraction (SS1017, SS1018)
We accept transformed bacterial cultures, grow them in our laboratory, and then extract the transformed plasmids using a plasmid prep kit prior to DNA sequencing. At the moment, we only accept stab culture for individual clones. 96-well format bacterial cultures are accepted as liquid cultures.
Ready-to-Load Sequencing with Clean-up (SS1019)
Customers may send us their pre-cycled sequencing reactions that are in single tubes or plate format. 1st BASE performs the post-cycle sequencing clean-up reaction for excess dye terminator removal and the subsequent analysis on our capillary electrophoretic analyzer.
DNA Sequencing Sample Quantification - By Agarose Gel (SS1020)
This additional quantification service provides customers with a means of verifying and quantifying their DNA sample prior to DNA sequencing. A small volume of DNA sample is run on an agarose gel to verify its purity and mass before proceeding with DNA Sequencing reactions.
DNA Sequencing Sample Quantification - Spectrophotometer Reading (SS1021)
This additional quantification service provides customers with a means of quantifying their DNA prior to DNA sequencing. A small volume of DNA sample is used to measure the OD260using the UV Spectrophotometer
